报告摘要:
Small interfering RNAs (siRNAs) and microRNAs (miRNAs) are essential gene regulators in eukaryotes. siRNAs are normally generated from double-strand RNAs, and miRNAs originate from miRNA precursors (pre-miRNAs) that fold back and form hairpin structures. It is typically expected one pre-miRNA to produce one miRNA. In recent studies by combining high-throughput deep sequencing, computational and experimental analyses, we discovered many pre-miRNAs in Arabidopsis that can produce siRNAs at the loci where miRNAs originate and multiple authentic miRNAs beyond cognate miRNAs. Many such novel miRNAs were expressed in response to stress, and many of these siRNAs were able to guide DNA methylation at the sites of their targets. Our analyses also showed that these novel miRNAs exist broadly in plants and animals and siRNA-producing pre-miRNAs exist also in rice and moss. Our results suggest that the pools of miRNAs and siRNAs are larger than were previously recognized, and small RNA-mediated gene regulation is broader and more complex than previously thought.
